SS-31 is a synthetic, mitochondria-targeted tetrapeptide (D-Arg-dimethylTyr-Lys-Phe-NH₂) belonging to the Szeto-Schiller peptide class. Its alternating aromatic-cationic structure allows it to cross the outer mitochondrial membrane and concentrate on the inner membrane independent of membrane potential, where its proposed mechanism centers on selective binding to cardiolipin — a phospholipid essential to cristae architecture and electron transport chain organization. In preclinical models it has been investigated for effects on cristae structure, electron transport efficiency, ATP production, and mitochondrial reactive oxygen species generation. It has been studied in research contexts spanning mitochondrial myopathy, ischemia-reperfusion injury, heart failure, and age-associated decline in muscle mitochondrial function. Also known as elamipretide, MTP-131, and Bendavia, it remains an investigational compound with no approved therapeutic indication.
